Letrozole is used to treat certain types of breast cancer in women who have already stopped menstruating (postmenopausal). It is also used for women who have already had other cancer treatments (eg, tamoxifen). This study suggests that there may be utility in increasing the dose even further, beyond 7.5 mg/day to as much as 12.5 mg/day.

- Anastrozole is an aromatase inhibitor, an anti-cancer medication use for the adjuvant or initial treatment of breast cancer in post-menopausal women.
- You should only take this medicine under strict medical supervision.
- If you’ve had an allergic reaction to letrozole or any of its ingredients, your doctor will likely not prescribe letrozole.
- Thus, there is reason to believe that higher-dose short-term administration may be more effective at inducing endogenous FSH release, resulting in greater follicular development.
- It should be used with caution in patients with liver problems due to alcohol to prevent the risk of liver damage.
And because Femara also lowers your estrogen level, the drug can add to this effect. This increases your risk of breaking a bone during Femara treatment. Letrozole is an aromatase inhibitor used in the treatment of breast cancer. Aromatase inhibitors work by inhibiting the action of the enzyme aromatase, which converts androgens into estrogens by a process called aromatization. As breast tissue is stimulated by estrogens, decreasing their production is a way of suppressing recurrence of the breast tumor tissue. Tamoxifen is also used to treat hormonally-responsive breast cancer, but it does so by interfering with the estrogen receptor.

Data derived from these patients suggested substantial inhibition of estradiol formation with doses of 2.5–5 mg daily. However, the application of these data to short-term use of the drug in reproductive age women is highly questionable. Nevertheless, clinical investigation of the drug in infertile women has been generally limited to 5 days of treatment at doses of 2.5–7.5 mg daily. Letrozol Denk (Letrozol Denk) is indicated for the adjuvant treatment of postmenopausal women with hormone receptor positive early breast cancer.
First and Second-Line Treatment of Advanced Breast Cancer
Be sure to always take the dosage of Femara that’s prescribed by your doctor. The usual dosage of Femara for inducing ovulation is 2.5 mg taken by mouth once daily for 5 days. Your doctor may ask you to start taking the drug either on day 3 or day 5 of your menstrual cycle. (Day 1 of your menstrual cycle is the first day of your period.) If you’re not having periods when starting Femara, your doctor will recommend when you should start taking the drug. To find out how effective Femara was for infertility in clinical studies, see the drug’s prescribing information.
- Femara has mainly been used to treat certain cases of breast cancer.
- Justification for not exceeding doses of 2.5–7.5 mg has been based on the concept that these doses reduce estradiol levels 88–98% [12].
- Specifically, your risk of bone fractures may increase after 5 years of taking this drug.
- Your fertility specialist will determine if letrozole is an appropriate treatment option for you based on your individual circumstances and medical history.
- For skeletal safety data from the adjuvant setting, please refer to Table 2.
The recommended dose is 2.5 mg once daily with or without food. Patients with severe liver impairment should receive 2.5 mg every other day. The effect of hepatic impairment on Letrozol Denk exposure in noncirrhotic cancer patients with elevated bilirubin levels has not been determined. Two meta-analyses of randomized, placebo-controlled studies suggest Letrozol Denk is superior to clomiphene for ovulation induction in subfertile females with polycystic ovary syndrome (PCOS). A greater increase in ovulation rate, clinical pregnancy rate, and live birth rate was observed with Letrozol Denk compared to clomiphene; complications were not found to differ between treatments. Additional trials may be necessary to further define the role of Letrozol Denk in this condition.
If you’re interested in finding an alternative to Femara, talk with your doctor. They can tell you about other medications that may work well for you. Femara can cause problems with fetal development (also called birth Sustanon 250 defects) and miscarriage if it’s taken during pregnancy.
